Do I need a music license to play music from my iTunes and Spotify playlists? If so, where would I get one?

Play music where? At home, no. In a public place, yes.

If you're playing in venues that regularly have DJs, bands and so on, they will in most cases have their own license for public performance though.

On the other hand, music from Spotify and similar services is for personal (home) use only. It's in their T&Cs.
 

Is it possible to get a one time use license for a wedding?
 

As I already said in my previous message, venues such as those typically used for weddings are normally licensed themselves, so it's not necessary for a DJ playing there to have a license (in the UK at least).

The above assumes that you're playing music that you have purchased - not streaming from Spotify (etc) or rented music.
