I have an event where i need to set up an active speaker in a separate room to disco, if i want to connect to my speaker from headphone socket of laptop via mini jack-phono how would i best set vdj sound settings to play through that. Just for some ceremony music (Speakers have xlr and phono inputs)

You should create a second "Master" entry on your audio setup and select the laptop's internal soundcard.
However I would not advise you to do that both for "software" and "hardware" reasons
For software, while it's possible to do it mixing different soundcards on your audio config is never a good idea and in some rare cases it may cause issues or introduce latency to the entire system.
For hardware, your laptop's headphones output was not designed to operate this way. Since the signal you will get from there is not balanced, running it on a long distance (bigger than 3-5 meters) most likely will introduce noise to the signal path. Also, the bigger the distance, the more the signal will fade out, which means that you'll have to increase the gain on the speaker itself. Increasing the gain will also result to increase the noise floor of the speaker e.t.c.
So generally it's NOT a good idea to go this way... It would be much much better to chain the speaker with the rest speakers using a balanced connection (XLR's) even if you have to run 50 or 100 meters of cable!

Hi, yes i think that may be a better option, although a bit difficult to see whats going on to cue the music. I may just set my backup controller/laptop up in the other room.

That is the better option. Even a mp3 player to speaker would be better than your first option.

In a pinch, a cell phone or tablet running DJAY or some other app works well. I have an iPad that I will load up with relevant tracks. I then put it into airplane mode and fire up DJAY 2. From here I go into an analog mixer to add in ceremony mics. If your just doing music, you can go directly to the speaker.

Use the "record/line out" on your mixer or amplifier ,to the amplifier of the other speaker.
